Copie directe in vitro d’un brin d’ADN par l’intermédiaire d’une polymérase bactérienne (isolée en 1988 d’un germe thermophile, Thermus aquaticus ) à température suffisamment élevée pour produire une réaction répétitive (autrement dit en chaîne). Cela permet l’amplification rapide, et aussi abondante qu’il est souhaité, du fragment moléculaire nucléique qui a servi d’amorce. La polymerase chain reaction constitue ainsi à la fois un agent biochimique précieux et un outil au service du diagnostic médical.
